唐高宗末年，高宗與武則天 (劉嘉玲-飾) 力排眾議出兵攻打敵國，豈料艦隊出海遭遇神秘事故，損失慘重，民間流傳艦隊出海時褻瀆了海中的神明龍王，故遇上不能解釋的怪事。年少的狄仁傑 (趙又廷-飾) 初到洛陽便捲入「花魁」銀睿姬 (Angelababy-飾) 遭擄走一案，狄仁傑幾經波折解救了睿姬，並獲得大理寺卿尉遲 ─ 真金 (馮紹峰-飾)的信任，遂聯同真金與醫術高明的沙陀忠 (林更新-飾) 一同查辦「龍王案」，找出神秘的海難之謎。睿姬被擄案，竟與其昔日情郎元鎮 (金汎-飾) 有關。睿姬原與富家公子元鎮相戀，但元鎮一日卻突然失蹤，從此下落不明。狄仁傑抽絲剝繭，漸漸發現元鎮的失蹤與海中的怪事有著莫大關係，甚至整個唐室的安危都受到威脅……
