柏加（Peter Parker）自小寄居在親友家中，一次偶然機會發現父親的遺物，遂夥同女友史黛茜（Gwen Stacy）尋找雙親離奇失踪的線索，並找上父親昔日的拍檔康博士（Dr. Connors）。一次意外地被蜘蛛咬傷後，柏加突然擁有如同蜘蛛般的超凡的攀爬能力；同時間，一隻巨型晰蜴突然在紐約四出破壞。柏加一方面要在女友及其 家人面前，隱藏「蜘蛛俠」的身份，另一方面又要承受大眾對「蜘蛛俠」的質疑；在晰蜴人橫行之際，柏加能否挺身而出，拯救萬千無辜市民？
